This quests starts with a woman who is in a small tent just above the arena:

The quest flag will send you to the following location but you can find the spiders all over the area so feel free to kill them while you do other quests.

NOTE: Some have gotten confused by this but it is not Tarantula Gravis spiders that you need to kill despite what the NPC tells you, it is actually Tarantula Medialis spiders that drop the Poison Glands.

Each Tarantula Medialis you kill will drop a Poison Gland when it dies:
